Description: BEND2 is a 799 amino acid protein that contains two BEN domains. BEND2 exists as two alternatively spliced isoforms and is considered a complete proteome. BEN domain mediates proteinDNA and proteinprotein interactions during chromatin organization and transcription. BEN domain may play a role in organization of viral DNA during replication or transcription. The BEND2 gene maps to human chromosome Xp22.13. Chromosome X consists of about 153 million base pairs and nearly 1,000 genes. The X and Y chromosomes are the human sex chromosomes. The combination of an X and Y chromosome lead to normal male development while two copies of X lead to normal female development. Color blindness, hemophilia, and Duchenne muscular dystrophy are well known X chromosome-linked conditions which affect males more frequently as males carry a single X chromosome.

Description: CP2 family gene products are reported to be involved in a-globin gene expression. Mouse CP2c (also known as a-CP2/CP2/LSF/UBP-1) is a homologue of human LBP-1c with 96% amino acid sequence identity. CP2c was initially identified as an activator of the mouse a-globin gene, which binds to the consensus DNA-binding sequence CNRG-N6-CNR(G/C) within a promoter element overlapping the CCAAT box. The transcriptional activity of CP2c was increased during induced differentiation of MEL cells and associated globin gene transcription. CP2c is also likely to be involved in regulation of non-globin erythroid-specific genes. It was reported that congenital erythropoietic porphyria was caused by the mutations in GATA-1 and CP2c binding sites within the promoter of the uroporphyrinogen III synthase gene, the fourth enzyme in the heme biosynthetic pathway.

Description: CALHM1 is a 346 amino acid multi-pass endoplasmic reticulum membrane protein that belongs to the FAM26 family. CALHM1 co-localizes with GRP 78 to the endoplasmic reticulum. Predominantly expressed in adult brain, CALHM1 may be a pore-forming ion channel that controls cytosolic Ca²⁺ permeability and cytosolic Ca²⁺ concentration in the cell. It is suggested that CALHM1 regulates amyloid precursor protein proteolysis and aggregated amyloid-beta peptides levels in a Ca²⁺ dependent manner. CALHM1 homomultimerizes and shares strong sequence similarities with the selectivity filter of the NMDA receptor, which generates a large Ca²⁺ conductance across the plasma membrane. CALHM1 may be a potential factor involved in the pathogenesis of Alzheimer's disease.

Description: NAP5 (Nck-associated protein 5), also known as peripheral clock protein, NCKAP5 or ERIH, is a 1,909 amino acid nuclear protein that is expressed in fetal and adult brain, leukocytes and fetal fibroblasts. Containing pro-rich sequences, NAP5 interacts with the adapter protein Nck via the SH3-containing region. Existing as four alternatively spliced isoforms, the gene encoding NAP5 maps to human chromosome 2q21.2 and mouse chromosome 1 E3. Human chromosome 2, the second largest human chromosome, consists of 237 million bases, encodes over 1,400 genes and makes up approximately 8% of the human genome. Harlequin icthyosis, a rare and morbid skin deformity, is associated with mutations in the ABCA12 gene present on chromosome 2. The lipid metabolic disorder sitosterolemia is associated with ABCG5 and ABCG8. An extremely rare recessive genetic disorder, Alstr syndrome, is due to mutations in the ALMS1 gene.

Description: Provides, together with MAGI1, an adhesion machinery at tight junctions, which may regulate the permeability of kidney glomerulus and small intestinal epithelial cells. Mediates calcium-independent homophilic cell adhesion. In testis, it may function as a cell adhesion molecule rather than a tight-junction protein. It may participate in the adhesion between spermatogonia-spermatogonia, spermatogonia-Sertoli cells, and Sertoli cells-Sertoli cells.

Description: The UNC5H family of proteins act as transmembrane receptors for netrin-1 and play a crucial role in axon guidance and migration of neural cells. In fact, UNC5H receptors express widely in cells that migrate, where they bind the G protein G Alpha 1-2 to inhibit G protein signaling. Additionally, UNC5H receptors induce apoptosis when cleaved by a caspase, producing an intracellular fragment containing a death domain, but this activity is blocked by the binding of netrin-1. The expression of UNC5H receptors is down-regulated in multiple cancers, including colorectal, breast, ovary, uterus, stomach, lung, and kidney cancers. Hence, in the absence of netrin-1, UNC5H receptors act as tumor suppressors by inhibiting anchorage-independent growth and invasion, but mutation of these receptors provides a potential mechanism for tumorigenicity. UNC5H2, also designated unc-5 homolog B or p53-regulated receptor for death and life protein 1 (p53RDL1) is highly expressed in brain with lower levels of expression observed in developing lung, cartilage, kidney and hematopoietic and immune tissues.

Description: Sox genes comprise a family of genes that are related to the mammalian sex determining gene SRY. These genes similarly contain sequences that encode for the HMG-box domain, which is responsible for the sequence-specific DNA-binding activity. Sox genes encode putative transcriptional regulators implicated in the decision of cell fates during development and the control of diverse developmental processes. The highly complex group of Sox genes cluster at least 40 different loci that rapidly diverged in various animal lineages. At present, 30 Sox genes have been identified. Members of this family have been shown to be conserved during evolution and to play key roles during animal development. Some are involved in human diseases, including sex reversal.

Description: Rsk1 is a member of a family of 90kDa ribosomal protein S6 kinases, which includes Rsk1, Rsk2 and Rsk3. These are broadly expressed serine/threonine protein kinases activated in response to mitogenic stimuli, including extracellular signal regulated protein kinases Erk1 and Erk2. Rsk1 is activated by MAPK in vitro and in vivo via phosphorylation. Active Rsks appear to play a major role in transcriptional regulation by translocating to the nucleus and phosphorylating c Fos and CREB.

Description: The TEK receptor tyrosine kinase is expressed almost exclusively in endothelial cells in mice, rats, and humans. This receptor possesses a unique extracellular domain containing 2 immunoglobulin-like loops separated by 3 epidermal growth factor-like repeats that are connected to 3 fibronectin type III-like repeats. The ligand for the receptor is angiopoietin-1. Defects in TEK are associated with inherited venous malformations; the TEK signaling pathway appears to be critical for endothelial cell-smooth muscle cell communication in venous morphogenesis.TEK is closely related to the TIE receptor tyrosine kinase.

Description: Representing about 2% of human DNA, chromosome 20 consists of approximately 63 million bases and 600 genes. Chromosome 20 contains a region with numerous genes expressed in the epididymis, which are thought important for seminal production, and some viewed as potential targets for male contraception. The PRNP gene encoding the prion protein associated with spongiform encephalopathies, like Creutzfeldt-Jakob disease, is found on chromosome 20. Amyotrophic lateral sclerosis, spinal muscular atrophy, ring chromosome 20 epilepsy syndrome and Alagille syndrome are also associated with chromosome 20. The C20orf160 gene product has been provisionally designated C20orf160 pending further characterization.
