Description: Mitogen-activated protein (MAP) kinase cascades are activated by various extracellular stimuli including growth factors. The MEK kinases (also designated MAP kinase kinase kinases, MKKKs, MAP3Ks or MEKKs) phosphorylate and thereby activate the MEKs (also called MAP kinase kinases or MKKs), including ERK, JNK and p38. These activated MEKs in turn phosphorylate and activate the MAP kinases. The MEK kinases include Raf-1, Raf-B, Mos, MEK kinase-1, MEK kinase-2, MEK kinase-3, MEK kinase-4, ASK 1 (MEK kinase-5) and MAP3K6 (MEK kinase-6). MEK kinase-1 has been shown to phosphorylate MEK-1 via Raf-independent pathway. Evidence suggests that MEK-3 is preferentially activated by MEK kinase-3 and that MEK-4 is activated by both MEK kinase-2 and MEK kinase-3. MEK kinase-4 has been shown to specifically activate the JNK path-way. ASK1 activates both MEK-4 and MEK-3/MEK-6 pathways.

Description: CacyBP is a 228 amino acid protein encoded by the human gene CACYBP. CacyBP is primarily a nuclear protein that contains one CS domain and one SGS domain. CacyBP is believed to be involved in calcium-dependent ubiquitination and subsequent proteosomal degradation of target proteins. It most likely serves as a molecular bridge in ubiquitin E3 complexes. It also participates in the ubiquitin-mediated degradation of b-catenin. CacyBP is thought to be a potential inhibitor of cell growth and invasion in the gastric cancer cell through its effects on b-catenin protein expression and transcriptional activation of TCF/LEF.

Description: Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. Its protein product is a protease present in seminal plasma. It is thought to function normally in the liquefaction of seminal coagulum, presumably by hydrolysis of the high molecular mass seminal vesicle protein. Serum level of this protein, called PSA in the clinical setting, is useful in the diagnosis and monitoring of prostatic carcinoma. Alternate splicing of this gene generates several transcript variants encoding different isoforms.

Description: MEK5 is a dual specificity protein kinase belonging to thr Ser/Thr protein kinase family, (MAP kinase kinase family). It is activated by phosphorylation on Ser/Thr by MAP kinase kinases and interacts specifically with ERK5, and not with another MAP kinase like P38. This kinase specifically interacts with and activates MAPK7/ERK5. This kinase itself can be phosphorylated and activated by MAP3K3/MEKK3, as well as by atypical protein kinase C isoforms (aPKCs). It is not phosphorylated by RAFA, RAFB or RAFC and it may interact with GTPases such as CDC42. The signal cascade mediated by this kinase is involved in growth factor stimulated cell proliferation and muscle cell differentiation. MEK5 is expressed in many adult tissues and is most abundant in heart and skeletal muscle.

Description: EphA3 is a member of the Ephrin (Eph) family. The Ephrins and Eph-related receptors comprise the largest subfamily of receptor protein-tyrosine kinases and have been implicated in mediating developmental events, especially in the nervous system and in erythropoiesis. Based on their structures and sequence relationships, ephrins are divided into the Ephrin A (EphA) class, which are anchored to the membrane by a glycosylphosphatidylinositol linkage, and the Ephrin B (EphB) class, which are transmembrane proteins. Receptors in the EPH subfamily typically have a single kinase domain and an extracellular region containing a Cys-rich domain and 2 fibronectin type III repeats.

Description: Ubiquitin-dependent proteolysis mediates selective destruction of various cell cycle regulators, transcription factors and tumor suppressors. In eukaryotic cells, selective breakdown of cellular proteins is ensured by their ubiquitination and subsequent degradation by the 26S proteasome. At specific stages of development, embryo- and tissue-specific components of the 26S proteasome form, facilitating proteolysis. 20S Proteasome ?, also designated macropain subunit C2 or PROS-30, is a prosomal protein involved in a non-lysosomal ATP/ubiquitin-dependent proteolytic pathway. The entire proteasome is composed of at least 15 non-identical subunits which form a highly-ordered ring-shaped structure.

Description: Encoding more than 700 genes, chromosome 15 is made up of approximately 106 million base pairs and is about 3% of the human genome. Angelman and Prader-Willi syndromes are associated with loss of function or deletion of genes in the 15q11-q13 region. In the case of Angelman syndrome, this loss is due to inactivity of the maternal 15q11-q13 encoded UBE3A gene in the brain by either chromosomal deletion or mutation. In cases of Prader-Willi syndrome, there is a partial or complete deletion of this region from the paternal copy of chromosome 15. Tay-Sachs disease is a lethal disorder associated with mutations of the HEXA gene, which is encoded by chromosome 15. Marfan syndrome is associated with chromosome 15 through the FBN1 gene. The FAM98 gene product has been provisionally designated FAM98 pending further characterisation.

Description: The human protooncogene JUN is the putative transforming gene of avian sarcoma virus 17, and it encodes a protein which is highly homologous to the viral protein. cJun (previously known as the Fos binding protein p39) and c Fos form a complex in the nucleus. AP 1 (activating protein 1) is a collective term referring to these dimeric transcription factors composed of Jun, Fos or ATF subunits that bind to a common DNA site, the AP1 binding site. AP 1 proteins, mostly the Jun group, regulate the expression and function of cell cycle regulators such as Cyclin D1, p53, p21 (cip1/waf1), p19 (ARF) and p16. Fos and Jun proto oncogene expression is induced transiently by a variety of extracellular stimuli associated with mitogenesis, differentiation processes or depolarisation of neurons. JUN has been mapped to 1p32 to p31, a chromosomal region involved in both translocations and deletions in human malignancies.

Description: Zinc-finger proteins contain DNA-binding domains and have a wide variety of functions, most of which encompass some form of transcriptional activation or repression. The majority of zinc-finger proteins contain a Krüppel-type DNA binding domain and a KRAB domain, which is thought to interact with KAP1, thereby recruiting histone modifying proteins. Zinc finger protein 575 (ZNF575) is a 245 amino acid member of the Krüppel C2H2-type zinc-finger protein family. Localized to the nucleus, ZNF575 contains six C2H2-type zinc fingers through which it is thought to be involved in DNA-binding and transcriptional regulation.

Description: GCF (GC-rich sequence DNA-binding factor), also known as C2orf3 (chromosome 2 open reading frame 3), transcription factor 9 (TCF-9) or DNABF, is a 781 amino acid nuclear protein that belongs to the GCF family. Widely expressed, GCF binds the GC-rich sequences of _-Actin, EGFR and calcium-dependent protease (CANP) promoters. GCF contains multiple phosphoserine and phosphothreonine residues, and two GCF isoforms are produced due to alternative splicing events. GCF is considered a candidate for susceptibility to dyslexia (DYX3) as both genes reside in close proximity on human chromosome 2. Chromosome 2 is the second largest human chromosome and consists of 237 million bases, encodes over 1,400 genes and makes up approximately 8% of the human genome.
