Description: Neuromedin U is a neuropeptide with high activity on smooth muscle. It is widely expressed in gastrointestinal systems and central nervous system (CNS). Peripheral activities of neuromedin U include smooth muscle stimulation, ion transport alterations in the gut and the regulation of local blood flow and adrenocortical function. Neuromedin U receptors 1 and 2 (NMUR1 and NMUR2) are multi-pass membrane proteins that belong to the G-protein coupled receptor 1 family of proteins. Both NMUR1 and NMUR2 act as receptors for the neuromedin U neuropeptide. NMUR1 is detected in peripheral organs, particularly in urogenital and gastrointestinal systems, with highest levels in testis. It’s expression in CNS is low, but the protein has been detected in cerebellum, hippocampus, dorsal root ganglia and spinal cord. NMUR2 is predominantly detected in central nervous system with highest levels detected in medulla oblongata, spinal cord and thalamus. It may also be detected in testis but has low levels of expression in peripheral tissues.

Description: The kit offers a set of six different MAbs specific for serine phosphorylation sites. Phosphorylation patterns in a given cell extract may differ when probed with different antibodies due to sequence specificity. Recognition therefore depends on two criteria: phosphorylation and surrounding aa motif. If one of the two criteria is not fulfilled, the antibody will not detect the phosphorylation site. Specificity has been determined by epitope mapping using degenerated phosphopeptide libraries. Clone 16B4 strongly interacts with pSK and pSP motifs, thus specifically recognizes substrates of MAP/SAP Kinases and CDC kinases.Clones 1C8, 4A3, 4A9, 4H4 and 7F12 interact with epitopes containing phosphoserine in the context of positive or neutral aa (e.g. substrates of PKA, PKB, PKC, PKG etc.) Though revealing a similar pattern in epitope mapping using phosphopeptide libraries, these antibodies interact with different sets of phosphoproteins in whole cell lysates.

Description: Dihydrorhodamine 123 is the reduced form of rhodamine 123, which is a commonly used fluorescent mitochondrial dye. It is non-fluorescent, but it readily enters most of the cells and is oxidized by oxidative species or by cellular redox systems to the fluorescent rhodamine 123 that accumulates in mitochondrial membranes. It is useful for detecting reactive oxygen species including superoxide (in the presence of peroxidase or cytochrome c) and peroxynitrite. Dihydrorhodamine 123 dihydrochloride is functionally equivalent to dihydrorhodamine 123 but with increased stability toward air oxidation and light during storage. Dihydroethidium (also called hydroethidium) is the chemically reduced form of the commonly used DNA dye ethidium bromide. The probe is useful to detect oxidative activities in viable cells, including respiratory burst in phagocytes. Dihydroethidium itself has blue fluorescence (355/420 nm) in cells, while the oxidized form ethidium has red fluorescence (518/605 nm) upon DNA intercalation. H2DCFDA (2′,7′-Dichlorodihydrofluorescein diacetate) is a useful fluorogenic reagent to detect reactive oxygen intermediates in cells. On oxidation, H2DCFDA becomes the highly green fluorescent 2′,7′-dichlorofluorescein, 504/529 nm (end product).
