Description: IL1RL1 (Interleukin 1 receptor-like 1), also known as ST2 and ST2V, is a member of the IL1R family and TLR superfamily. TIR proteins initiate the innate immune response through NF-kB. Unlike other TIR proteins, IL1RL1 doesn't activate NF-kB. Instead, it promotes endotoxin tolerance through suppressive interaction with TLR4 and IL1R. This TLR inhibition also promotes a Th2 response.

Description: Granulocyte-colony stimulating factor (G-CSF) is a cytokine that functions as a potent inducer of neutrophilic granulocyte proliferation, terminal differentiation, and activation. G-CSF synthesis occurs in monocyte, macrophage, epithelial, endothelial, and fibroblast cells after activation by bacterial endotoxins, tumor necrosis factor alpha (TNF-α), interleukin 1 (IL-1), or interleukin 17 (IL-17). The functional activity of G-CSF is mediated through the granulocyte colony-stimulating factor receptor (G-CSF-R) to activate JAK/STAT and MAPK signal transduction pathways. G-CSF also promotes neurogenesis and inhibits neuronal apoptosis. Human and mouse G-CSF proteins are cross-reactive.

Description: IDO catalyzes the first and rate-limiting step in the main pathway of human tryptophan catabolism, the kynurenine pathway. Proinflammatory mediators, such as endotoxin and IFN-gamma induce the expression of IDO in several tissues. IDO-dependent suppression of T-cell responses might function as natural immunoregulatory mechanism. Physiological IDO activity has been implicated in T-cell tolerance to tumors, dysfunctional selftolerance in non-obese diabetic (NOD) mice, and as a protective negative regulator in autoimmune disorders.

Description: Mouse Interleukin-1 (IL-1) designates two proteins, IL-1 alpha and IL-1 beta , which are the products of distinct genes, but recognize the same cell surface receptors. IL-1 alpha and IL-1 beta are structurally related polypeptides that show approximately 25% homology at the amino acid level. Both proteins are produced by a wide variety of cells in response to stimuli such as those produced by inflammatory agents, infections, or microbial endotoxins. The proteins are synthesized as 31 kDa precursors that are subsequently cleaved into proteins with molecular weights of approximately 17.5 kDa.

Description: ST2 Antibody: ST2 is a member of a superfamily containing the interleukin-1 receptor and the Toll-like receptors (TLRs). The TLRs are signaling molecules that recognize different microbial products during infection and serve as an important link between the innate and adaptive immune responses. ST2 was originally identified as a protein whose production was stimulated by various proliferation-inducing agents such as PDGF and FGF. More recently, it has been shown to negatively regulate IL-1 receptor and Toll-like receptor (TLR) 4 signaling and to maintain endotoxin tolerance. It has been suggested that the inhibition of TLR4 signaling occurs through the association and sequestering of TLR adaptor molecules such as MyD88 and TIRAP.

Description: Heme Oxygenase 1 (HO-1) is an enzyme in endoplasmic reticulum that belongs to the heme oxygenase family. HO-1 cleaves the heme ring at the alpha methene bridge to form Biliverdin. Biliverdin is subsequently converted to Bilirubin by Biliverdin reductase. In physiological state, the highest activity of HO-1 is found in the spleen, where senescent erythrocytes are sequestrated and destroyed. HO-1 activity is highly inducible by its substrate heme and by various non-heme substances such as heavy metals, bromobenzene, endotoxin, oxidizing agents and UVA. HO-1 is involved in the regulation of cardiovascular function and response to a variety of stressors. Defects in HO-1 are the cause of Heme Oxygenase 1 deficiency, resulting in marked erythrocyte fragmentation and intravascular hemolysis, coagulation abnormalities, endothelial damage, and iron deposition in renal and hepatic tissues.

Description: Caspases are cysteine proteases that cleave C-terminal aspartic acid residues on their substrate molecules. This gene is most highly related to members of the ICE subfamily of caspases that process inflammatory cytokines. In rodents, the homolog of this gene mediates apoptosis in response to endoplasmic reticulum stress. However, in humans this gene contains a polymorphism for the presence or absence of a premature stop codon. The majority of human individuals have the premature stop codon and produce a truncated non-functional protein. The read-through codon occurs primarily in individuals of African descent and carriers have endotoxin hypo-responsiveness and an increased susceptibility to severe sepsis. Several alternatively spliced transcript variants have been noted for this gene.

Description: Cathelicidin Antibody: One component of host defense at mucosal surfaces is epithelial-derived antimicrobial peptides. Cathelicidins are one family of antimicrobial peptides characterized by conserved pro-peptide sequences that have been identified in epithelial tissues and some myeloid cells of humans and animals. LL-37/hCAP-18 is the only Cathelicidin found in humans and is expressed in inflammatory and epithelial cells. The presence of these molecules is essential for defense against invasive bacterial infection in skin. Besides their direct antimicrobial function, Cathelicidins have multiple roles in mediating innate and adaptive immunity, such as endotoxin neutralizing, angiogenesis, wound healing and promoting neutrophil chemotaxis and mast cell recruitment. Finally, Cathelicidin antimicrobial peptides qualify as prototypes of innovative drugs that may be used to treat infection and/or modulate the immune response.

Description: Use visfatin to catalyse the production of nicotinamide mononucleotide from nicotinamide (Revollo <i>et al.</i>). Nicotinamide mononucleotide is a precursor to nicotinamide adenine dinucleotide (NAD) that is vital to energy metabolism, cell death, and other cellular processes (Ying). Visfatin also acts as an immunomodulator by activating leukocytes and inducing the production of pro-inflammatory and anti-inflammatory cytokines (Moschen <i>et al.</i>), and there is evidence that visfatin can regulate insulin receptor signaling and insulin secretion (Brown <i>et al.</i>). Belonging to the nicotinate phosphoribosyltransferase (NAPRTase) family, Visfatin is an adipokine that is produced in adipocytes, leukocytes, and hepatocytes (Chiu <i>et al.</i>, Garten <i>et al.</i>, Kralisch <i>et al.</i>). It is active as a homodimer with each identical monomer consisting of two structural domains made up of 19 β-strands and 13 α-helices (Kim <i>et al.</i>). For consistency and reproducibility across your applications, visfatin from STEMCELL comes lyophilised with ≥90% purity, and is verified by LAL analysis to ensure endotoxin levels are ≤1,0 EU/μg protein.

Description: A member of the pentraxin family of proteins, C-reactive protein (CRP) plays important roles in host defenses, including regulating complement activation in vivo (Sjoberg <i>et al.</i>), decreasing nitric oxide production, and inhibiting angiogenesis in vitro (Verma <i>et al.</i>, 2002). At specific concentrations, CRP directly inhibits endothelial progenitor cell (EPC) differentiation, survival, and function, and is thus implicated in the development of cardiovascular diseases (Verma <i>et al.</i>, 2004). CRP is primarily synthesized by liver hepatocytes, but also by other immune cells, such as macrophages and lymphocytes (Sproston and Ashworth). The expression of this acute-phase protein is induced by IL-6 and IL-1 secretion of macrophages and T-cells during inflammation. For consistency and reproducibility across your applications, C-reactive protein from STEMCELL comes lyophilised with ≥87% purity, and endotoxin levels are verified to be ≤1,0 EU/μg protein.

Description: Use resistin to modulate various cellular responses, such as promoting neutrophil extracellular trap (NET) formation (Jiang <i>et al.</i>) and regulating anti-inflammatory signaling pathways through toll-like receptor 4 (TLR4) (Jang <i>et al.</i>, 2017). A member of the resistin-like molecule (RELM) family, resistin is produced by adipocytes in mice, and has been implicated in insulin resistance, reducing glucose tolerance, and insulin sensitivity <i>in vivo</i> (Li <i>et al.</i>). In humans, resistin is expressed predominantly in leukocytes, modulating inflammation in numerous diseases (Jamaluddin <i>et al.</i>; Jang <i>et al.</i>, 2015; Mantula <i>et al.</i>). Studies also show that resistin facilitates vascular endothelial growth factor (VEGF)-A-dependent angiogenesis in human chondrosarcoma cells, highlighting it as a promising target for chondrosarcoma angiogenesis (Chen <i>et al.</i>). For consistency and reproducibility across your applications, resistin from STEMCELL comes lyophilised with ≥95% purity, and endotoxin levels are verified to be ≤1,0 EU/μg protein.

Description: Commonly known as vaspin, Serpin A12 is an adipokine that is thought to perform insulin-sensitizing functions, such as the regulation of kallikrein 7 (KLK7), an insulin-inhibiting protease (Heiker <i>et al.</i>). The Serpin A12 glycoprotein belongs to the serine protease inhibitor family (serpin), and is mainly associated with obesity, insulin resistance, and glucose metabolism, but may also protect against high fat-induced bone loss (Wang <i>et al.</i>). Serpins share a highly conserved core structure with multiple α-helices, β-strands, and a reactive site loop (RCL) to interact with the target protease (Huntington). It is expressed predominantly in visceral adipose tissue and at significantly higher amounts in obese individuals (Kurowska <i>et al.</i>). This protein contains a His-residue tag at the carboxyl end of the polypeptide chain. For consistency and reproducibility across your applications, Serpin A12 from STEMCELL comes lyophilised with ≥94% purity, and is verified by LAL analysis to ensure endotoxin levels are ≤1,0 EU/μg protein.
