Description: Agar is a polysaccharide complex extracted by bleaching and hot water treatment of agarocytes from the red alga Rhodophyceae, and usually consisting of the genera Gelidium, Acanthopeltis, Ceramium, Pterocladia and Gracilaria. The algae are typically found in the Pacific and Indian Oceans and in the Sea of Japan. It is primarily composed of two different units: Agarose and Agaropectin; Agarose is a neutral gelling component which is composed of a linear polymer of alternating D-galactose and 3,6-anhydrogalactose units. Agaropectin is a non-gelling component which consists of D-1,3-glycosidically linked D-galactose units, some of which are sulfated at the 6th position.

Description: EC Blue is a simple, quick and reliable test method for the qualitative and quantitative determination of coliforms and <i>E.coli</i> in water. Add water to be tested into a pre-prepared, sterile, non-fluorescent EC Blue container. Then incubate the mixture at 35 ±2 °C. The medium does not require further preparation work. This means that the test can be carried out anywhere without much effort. Traditional incubating methods require about three days for the detection of coliforms or <i>E.coli</i>. EC Blue, on the other hand, already provides reliable test results after 24 hours. The detection is based on a colour reaction and is thus easy to interpret: if coliform bacteria are present, the water turns blue due to the cleavage of the chromogenic substrate X-Gal. In the event of faecal contamination by <i>E.coli</i>, these bacteria can only be conclusively detected under UV light (reaction with a fluorescent dye). Suspended matter, cloudy, yellowish or chlorinated water samples do not generally interfere with detection. EC Blue has a shelf life of up to two years at room temperature if protected from light.

Description: M-Endo broth is used for the estimation of coliform bacteria in water samples using the membrane filtration technique. It is possible to remove bacteria from fluids by passing them through filters with such small pore size that bacteria are arrested. This filtration technique enables fairly large volumes of water to pass rapidly under pressure, but prevents the passage of any bacteria present. These nutrients are retained on the surface of the membrane which is then brought into contact with suitable liquid nutrients. These diffuse upwards through the pores thereby inducing the organisms to grow as surface colonies which can be counted.

For public wastewater and drinking water: MF-Endo and M-FC (with and without rosolic acid for helping to increase the specificity of the medium for faecal coliforms) media are used when analysing water samples for total coliforms, faecal coliforms and <i>E. coli.</i> For food and beverage: M-TGE, HPC, M-Green YM, and orange serum broths are used in the food and beverage industry to QC final products (and fluids used in the manufacture of these products) to detect potential spoilage organisms. Trypticase soy broth, M-TGE, and HPC may be used to test final products and process waters for total bacteria in the pharmaceutical and microelectronics industries. <i>Pseudomonas </i>and <i>KF-Streptococcal</i> broths are highly selective media for organisms in these genera, and are used to monitor samples wherever contamination by these organisms must be prevented.
