Description: Retinoids are metabolites of vitamin A (retinal) and are believed to represent important signaling molecules during vertebrate development and tissue differentiation. Two families of retinoid receptors have been identified. Retinoic acid receptors (RARs) include RAR alpha, RAR beta and RAR gamma, each of which has a high affinity for all trans retinoic acids and belongs to the same class of nuclear transcription factors as thyroid hormone receptors, vitamin D3 receptor and ecdysone receptor. The ligand binding domains of the RARs are highly conserved and RAR isoforms are expressed in distinct patterns through out development and in the mature organism. Members of the retinoid X receptor (RXR) family, RXR alpha, RXR beta and RXR gamma, are activated by 9 cis retinoic acid, a stereo and photoisomer of all trans RA that is expressed in vivo in both liver and kidney and may represent a widely used hormone.

Description: The NGG1 interacting factor 3-like 1 (NIF3L1) is a 377 amino acid protein expressed mainly in the cytoplasm of cells in several different tissues. It has been highly conserved throughout evolution, from bacteria to mammals. NIF3L participates in retinoic acid-primed neural differentiation of P19 embryonic carcinoma cells by cooperating with Trip15/CSN2, a transcriptional corepressor/component of COP9 signalosome. NIF3L1 interacts with itself and with the NIF3L1 binding protein 1 (NIF3L1 BP1), which is a novel protein presumed to contain a leucine zipper domain.

Description: Adapalene, a synthetic naphthoic acid derivative, is a selective agonist of retinoic acid receptor (RAR)-β and RAR-γ (Kds = 34 and 130 nM, respectively). At 0,1-10 µM, adapalene dose-dependently inhibits sebocyte growth, proliferation and differentiation in a primary rat preputial cell culture system. Adapalene demonstrates anti-inflammatory and immunomodulatory activity by inhibiting 5-lipoxygenase-catalyzed leukotriene production and expression of toll-like receptor 2 on human monocytes. It is commonly used for the study of the pathogenesis and treatment of acne vulgaris.

Description: P2Y purinoceptor 11 (P2Y11) is a 374 amino acid protein belonging to the G-protein coupled receptor one family. P2Y11 is a multi-pass cell membrane protein that acts as a receptor for both ATP and ATD coupled to G proteins. Due to these interactions, P2Y11 is involved in phosphatidylinositol-calcium and adenylyl cyclase pathways. Induced by DMSO and retinoic acid, P2Y11 is highly expressed in spleen tissue. A putative trans-splicing event involving the gene that encodes P2Y11 and an upstream gene encoding PPAN has been found to result in a fusion protein, designated PPAN-P2RY11.

Description: 3-alpha-hydroxysteroid dehydrogenase that converts 3-alpha-tetrahydroprogesterone (allopregnanolone) to dihydroxyprogesterone and 3-alpha-androstanediol to dihydroxyprogesterone. May play a role in the biosynthesis of retinoic acid from retinaldehyde, but seems to have low activity with retinoids. Can utilize both NADH and NADPH.Tissue specificity:Highly expressed in trachea and epidermis. Detected at lower levels in spinal cord, bone marrow, brain, tongue, esophagus, heart, colon, testis, placenta, lung, skeletal muscle and lymph node.

Description: PCGF1 is a component of the Polycomb group (PcG) multiprotein BCOR complex, a complex required to maintain the transcriptionally repressive state of some genes, such as BCL6 and the cyclin-dependent kinase inhibitor, CDKN1A. It represses CDKN1A expression by binding to its promoter, and this repression is dependent on the retinoic acid response element (RARE element). PCGF1 als promotes cell cycle progression and also enhances cell proliferation, thus it may have a positive role in tumor cell growth.

Description: The steroid receptor superfamily acts through direct association with DNA sequences known as hormone response elements (HREs) and binds DNA as either homo- or heterodimers. The promiscuous mediator of heterodimerization, RXR, is the receptor for 9-cis retinoic acid, and dimerizes with VDR, TR, PPAR, and several novel receptors including LXR (also referred to as RLD-1) and FXR. FXR and LXR fall into a category of proteins termed “orphan receptors” because of their lack of a defined function, and in the case of LXR, the lack of a defined ligand. FXR has been shown to bind a class of lipid molecules called farnesoids. LXR/RXR heterodimers have highest affinity for DR-4 DNA elements while FXR/RXR heterodimers bind IR-1 elements. Both LXR/RXR and FXR/RXR heterodimers retain their responsiveness to 9-cis retinoic acid.

Description: DDAH, a dimethylarginine dimethylaminohydrolase, hydrolyses dimethyl arginine (ADMA) and monomethyl arginine (MMA), both inhibitors of nitric oxide synthases, and may be involved in in-vivo modulation of nitric oxide production. Impairment of DDAH causes ADMA accumulation and a reduction in cGMP generation. DDAH II, the predominant DDAH isoform in endothelial cells, facilitates the induction of nitric oxide synthesis by all-trans-Retinoic acid (atRA) (4). DDAH proteins are highly expressed in colon, kidney, stomach and liver tissues.
