Description: TRAIL-R1 is a receptor for the cytotoxic ligand TRAIL. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. Promotes the activation of NF-kappaB.

Description: CCND3 encodes a protein that belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance through the cell cycle. Cyclins function as regulators of CDK kinases. Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event. This cyclin forms a complex with and functions as a regulatory subunit of CDK4 or CDK6, whose activtiy is required for cell cycle G1/S transition. This protein has been shown to interact with and be involved in the phosphorylation of tumor suppressor protein Rb. The CDK4 activity associated with this cyclin was reported to be necessary for cell cycle progression through G2 phase into mitosis after UV radiation.

Description: The protein encoded by CDK8 is a member of the cyclin-dependent protein kinase (CDK) family. CDK family members are highly similar to the gene products of Saccharomyces cerevisiae cdc28, and Schizosaccharomyces pombe cdc2, and are known to be important regulators of cell cycle progression. This kinase and its regulatory subunit cyclin C are components of the RNA polymerase II holoenzyme complex, which phosphorylates the carboxy-terminal domain (CTD) of the largest subunit of RNA polymerase II. This kinase has also been shown to regulate transcription by targeting the CDK7/cyclin H subunits of the general transcription initiation factor IIH (TFIIH), thus providing a link between the 'Mediator-like' protein complexes and the basal transcription machinery.

Description: ROC1 encodes an evolutionarily conserved protein that interacts with cullins. The protein plays a unique role in the ubiquitination reaction by heterodimerizing with cullin-1 to catalyze ubiquitin polymerization. It also may be involved in the regulation of protein turn-over.

Description: MDM4 inhibits p53- and p73-mediated cell cycle arrest and apoptosis by binding its transcriptional activation domain. It inhibits degradation of MDM2. It can reverse MDM2-targeted degradation of p53 while maintaining suppression of p53 transactivation and apoptotic functions. The human MDM4 gene, which plays a role in apoptosis, encodes a 490-amino acid protein containing a RING finger domain and a putative nuclear localization signal. The MDM4 putative nuclear localization signal, which all Mdm proteins contain, is located in the C-terminal region of the protein. The mRNA is expressed at a high level in thymus and at lower levels in all other tissues tested. MDM4 protein produced by in vitro translation interacts with p53 via a binding domain located in the N-terminal region of the MDM4 protein. MDM4 shows significant structural similarity to p53-binding protein MDM2. Publication Note: This RefSeq record includes a subset of the publications that are available for this gene. Please see the Entrez Gene record to access additional publications.

Description: Lymphocyte activation gene 3 (LAG-3; CD223) plays an important role in negatively regulating T cell proliferation, function and homeostasis. It is required for maximal natural and induced regulatory T cell (Treg) function. LAG-3 is closely related to the T cell co-receptor CD4 and binds to MHC class II molecules but with a significantly higher affinity than CD4.

Description: The LTbetaR activates two different NF-kappaB pathways that lead to distinct patterns of gene induction, including selected chemokines and the cytokine BAFF, which is essential for the survival of mature B lymphocytes. LTbetaR activates the classical NF-kappaB (relA/p50) pathway, like the type 1 TNF receptor (TNFR1), that regulates proinflammatory genes, like the chemokine MIP1beta. However, LTbetaR, unlike TNFR1, also activates the processing of p100 to form RelB/p52 complexes, which activate genes involved in lymphoid organ formation and lymphocyte survival.

Description: RIG-I and MDA5 are highly conserved helicases involved in the innate immune response to virus. RIG-I is a member of the DEAD-box RNA helicases and is activated by cytoplasmic dsRNA and 5’-ppp RNA produced during the viral replication. The protein is characterized by a N-terminal region with two caspase recruitment domains (CARD) and a C-terminal region harboring potential ATP-dependent RNA helicase activity. RIG-I recruits the CARD adaptor inducing IFN-beta (Cardif) in a CARD-CARD-dependent manner resulting in NF-kappaB and IRF3 activation.

Description: IFN gamma receptor beta is part of the receptor for interferon gamma. This class II cytokine receptor pairs with CDw119 to form the IFN gamma receptor and is an integral part of the IFN gamma signal transduction pathway. CDw119 serves as the IFN gamma binding chain and associates with the IFN gamma beta chain which is required for receptor signaling. The extracellular portion of both the IFN gamma receptor alpha and beta chains must be species matched. The IFN gamma receptor beta chain is expressed on T and B cells, NK cells, monocytes/ macrophages, and fibroblasts. Binding of IFN gamma induces receptor dimerization, internalization, Jak1 and Jak2 protein kinase activation and, ultimately, STAT1 activation. It is also likely to interact with GAF. IFN gamma initiates and regulates a variety of immune responses and is required for signal transduction. Contains 2 fibronectin type III domains. Defects in IFN gamma Receptor beta are a cause of mendelian susceptibility to mycobacterial disease (MSMD), a rare condition that confers predisposition to illness caused by several mycobacteria strains

Description: CES1 is one of the enzymes responsible for the hydrolysis of ester- and amide-bond-containing drugs such as cocaine and heroin. They also hydrolize long-chain fatty acid esters and thioesters. This enzyme is known to hydrolyze aromatic and aliphatic esters and is necessary for cellular cholesterol esterification. It may also play a role in detoxification in the lung and/or protection of the central nervous system from ester or amide compounds. Carboxylesterase deficiency may be associated with non-Hodgkin lymphoma or B-cell lymphocytic leukemia.Carboxylesterase 1 is a member of a large multigene family.
