Description: Stimulating T cells with HHV8 (K8) peptide pool releases downstream cytokines and upregulates activation markers, enabling antigen-specific T cells to be detected or isolated for analysis. HHV8 (K8) peptide pool is a lyophilised mixture of 57 peptides from protein K8 alpha of human herpesvirus 8 (HHV-8), and consists of 15-mer peptides with 11-amino-acid overlaps that cover amino acids 1 to 237 on K8. A basic leucine zipper (bZip) protein that associates with the lytic origin of HHV8 (Lin <i>et al.</i>), K8 is thought to play a role in lytic DNA replication, as a trans-acting factor (AuCoin <i>et al.</i>). It has also been found to modulate the activity of the transcriptional transactivator K-Rta (Izumiya <i>et al.</i>), and has an autoregulatory function (Wang <i>et al.</i>). Viral peptide pools are useful for a broad range of applications, including vaccine development, immunological research, and diagnostic assay development.

TNF-α has cytotoxic effects on cancer cells in vitro by stimulating anti-tumor immunosuppressive responses. TNF-α stimulates expression of E- and P-selectins, thus facilitating adhesion of neutrophils, monocytes, and memory T cells to activated platelets and endothelial cells (Zelová andamp; Hosek). Other effects of TNF-α include vasodilatation and edema formation.

Description: Interleukin 12 (IL-12p70) is a heterodimeric cytokine composed of p35 and p40 subunits. The receptor is also expressed on basophils, eosinophils, mast cells, endothelial cells, fibroblasts, monocytes, macrophages, respiratory epithelial cells, and smooth muscle cells (Hershey). Signaling through the IL-13 receptor activates the JAK/STAT and IRS-1/IRS-2 pathways. <i>in vivo</i>, IL-13 has a role in resistance to extracellular helminth parasites by regulating gastrointestinal parasite expulsion, as well as in airway hyperresponsiveness, allergic inflammation, tissue remodeling, tumor cell growth, and fibrosis (Wynn 2015). Secreted IL-13 is a protein consisting of 112 amino acids with a molecular mass of 10 kDa (Hershey). Human IL-13 is not species-specific but has greater activity on human cells compared to mouse cells (Hershey).
